Proposed mechanisms include brain-derived neurotrophic factor (BDNF) modulation and increased dopamine receptor density in animal studies. Its short half-life of approximately 30 minutes has led to intranasal administration protocols designed to bypass first-pass metabolism and achieve central nervous system exposure. Clinical data are predominantly from Russian-language publications. Individuals tracking semax should document dosing protocols, cognitive effects, and any neurological symptoms.
Mechanism
A synthetic peptide related to a fragment of ACTH(4-10) with the melanocortin activity largely removed. In preclinical research it has been associated with neuroprotective and cognitive effects, possibly involving BDNF signaling; the mechanism in humans is not well characterized.
Research summary
Most research originates from a limited number of groups, largely preclinical or small studies, primarily in Russia where a related formulation has been used. It is not an FDA-approved drug, and independent modern clinical data are limited. Claims should be read against that evidence gap. In July 2026 an FDA advisory committee (the Pharmacy Compounding Advisory Committee) voted to recommend this compound for the 503A bulk-compounding list. That recommendation is non-binding and is not FDA approval — it would still require formal rulemaking, and no approved clinical evidence of safety or efficacy exists.
