Ipamorelin is a synthetic pentapeptide that acts as a selective ghrelin receptor (GHS-R1a) agonist, stimulating growth hormone (GH) release from the pituitary. Unlike earlier GH secretagogues such as GHRP-6, ipamorelin shows minimal effect on cortisol and prolactin in early studies. Its short half-life of approximately 2 hours typically leads to protocols involving once- or twice-daily subcutaneous administration. Ipamorelin is frequently combined with a GHRH analogue (such as Mod GRF 1-29 or CJC-1295) to produce a synergistic GH pulse. Tracking dose timing relative to meals (GH secretion is suppressed by hyperglycaemia), injection site rotation, and any side effects is essential for evaluating response.
