Liraglutide is a once-daily GLP-1 receptor agonist whose fatty-acid side chain enables albumin binding sufficient for a 13-hour half-life but not once-weekly dosing. It was the first GLP-1 RA approved for weight management (Saxenda) and remains widely used for type 2 diabetes (Victoza). Its shorter half-life means daily injection is required, which can be an advantage for patients who prefer tighter control over drug exposure or who experience side effects with longer-acting agents. Common injection sites are the abdomen, thigh, and upper arm. Tracking daily doses, nausea patterns, and weight trends helps patients and clinicians find the right dose schedule.

Mechanism

A GLP-1 receptor agonist with a fatty-acid acylation that promotes albumin binding and extends its half-life to allow once-daily dosing. Like other GLP-1 agonists it is associated with glucose-dependent insulin secretion, slowed gastric emptying, and reduced appetite.

Research summary

Approved for type 2 diabetes (LEAD program; cardiovascular outcomes in LEADER) and, at a higher dose, for chronic weight management (SCALE program). Marketed as Victoza for diabetes and Saxenda for weight management.

Dosing in the literature

Trials evaluated once-daily subcutaneous administration with titration. Specific regimens are individualized and set by a prescribing clinician; they are outside the scope of this summary.

Reported side effects

Gastrointestinal effects (nausea, vomiting, diarrhea) are most commonly reported. Prescribing information includes warnings such as thyroid C-cell tumor risk in rodents and pancreatitis. This is not a complete safety profile.

Stacking notes

Combination with insulin or sulfonylureas raises hypoglycemia considerations and is a clinical decision.
