Reconstituting a lyophilised (freeze-dried) peptide for the first time can feel intimidating. Between the mg per vial, the mL of BAC water, and the syringe units, there is plenty of room for error. This guide breaks down the process into clear steps.

What you need

  • A vial of lyophilised peptide (e.g. 10 mg)
  • Bacteriostatic water (BAC water)
  • An alcohol swab
  • A U-100 insulin syringe
  • A reconstitution calculator (or pen and paper)

Step 1: Check your vial

Every peptide vial should list the total amount of peptide in milligrams (mg). Common values are 2 mg, 5 mg, 10 mg, and 15 mg. If the label is unclear, do not guess — contact your supplier.

Step 2: Choose your BAC water volume

Most peptides are reconstituted with 1–3 mL of BAC water. A good rule of thumb: use 1 mL for small vials (2–5 mg) and 2 mL for 10 mg vials. The more BAC water you use, the more syringe volume you will need to draw for a given dose.

Step 3: Calculate concentration

Concentration (mg/mL) = Total peptide (mg) / BAC water (mL)

Example: 10 mg vial + 2 mL BAC water = 5 mg/mL

Step 4: Calculate your dose

Dose volume (mL) = Desired dose (mg) / Concentration (mg/mL)

On a U-100 syringe, 1 mL = 100 units.

Example: Want 0.5 mg from a 5 mg/mL solution → 0.1 mL = 10 units.

Step 5: Inject safely

  1. Swab the vial stopper with alcohol.
  2. Inject BAC water slowly, aiming the stream at the glass wall (not directly at the peptide powder).
  3. Swirl gently — do not shake.
  4. Let it sit for a few minutes until fully dissolved.
  5. Draw your dose into the syringe.
  6. Choose a rotation site (abdomen, thigh, upper arm) and rotate each time.
