> PLACEHOLDER — pending clinical review by licensed pharmacist. Dulaglutide is a once-weekly GLP-1 receptor agonist engineered as an Fc-fusion protein. By fusing a GLP-1 analogue to a modified IgG4 Fc fragment, the molecule exploits the neonatal Fc receptor (FcRn) recycling pathway to achieve a half-life of approximately 120 hours. It is marketed as Trulicity for type 2 diabetes. Compared with other once-weekly GLP-1 RAs, dulaglutide has a distinct dosing pen and titration schedule. Patients tracking their therapy in a log app can benefit from recording dose timing, injection site rotation, and gastrointestinal side effects to identify personal tolerance patterns.

Mechanism

A long-acting GLP-1 receptor agonist in which the GLP-1 peptide is fused to an antibody Fc fragment, extending its half-life to allow once-weekly dosing. Like other GLP-1 agonists it is associated with glucose-dependent insulin secretion, slowed gastric emptying, and reduced appetite.

Approved for type 2 diabetes and studied in the AWARD program, including a cardiovascular outcomes trial (REWIND). Marketed as Trulicity.

Reported side effects

Gastrointestinal effects (nausea, diarrhea, vomiting) are most commonly reported. Prescribing information includes warnings such as thyroid C-cell tumor risk in rodents and pancreatitis. This is not a complete safety profile.

Stacking notes

Combination with other glucose-lowering agents (especially insulin or sulfonylureas) raises hypoglycemia considerations and is a clinical decision.
